Influence of Autologous Bone Marrow Mesenchymal Stem Cells Transplantation by Intracoronary Infusion on Angiogenesis and Cardiac Function in Myocardial Infarction Rabbits

Abstract

Objective To observe the influence of autologous bone marrow mesenchymal stem cells(BM-MSCs)transplantation by intracoronary infusion on angiogenesis and cardiac function in myocardial infarction rabbits.Methods Forty-five white Japanese big-ear rabbits were divided randomly into 3 groups,BM-MSCs,control and sham operation,15 in each.In BM-MSCs group,bone marrow was extracted to establish AMI models by separating,purifying and in vitro amplifying BM-MSCs,and ligating left anterior descending(LAD)of coronary artery,and 5-bromodeoxyuridine(BrdU,5×106)labeled BM-MSCs was infused into LAD after reperfusion.In control,AMI models were established in the same way,infusing equivalent serum-free medium.And in sham operation group the same things were done except that LAD was not ligated.24 h or 4 weeks after operation,left ventricle ejection fraction(LVEF)and fractional shortening(FS)of the ventricular minor-semi axis were measured by echocardiography.after 4 weeks,the survival of transplanted BM-MSCs was evaluated by BrdU antibody immunohistochemical staining,and the expression of vascular endothelial growth factor(VEGF)protein and the changes of capillary density were detected in all groups.Results Twenty-four hours after transplantation,LVEF and FS of sham operation group were better than those of groups BM-MSCs and control(P0.01).Four weeks after,LVEF and FS of groups BM-MSCs and sham operation were better than those of control group(P0.01),and anti-BrdU antibody immunohistochemistry showed positive in BM-MSCs group.The expression of VEGF and capillary density were more abundant in BM-MSCs group than in control and sham operation(P0.01).Conclusion Autologus BM-MSCs transplanted by intracoronary infusion can survive in AMI region,augment expression of VEGF protein and induce angiogenesis,thus to improve cadiac function.
