Optimization of Cultivation Conditions and Properties of Laccase from White Rot Fungus Trametes pubescens MB89

Abstract

The optimal fermentation conditions for laccase production by white-rot fungi Trametes pubescens MB89 were studied and laccase properties were analyzed.Culture methods,temperature and copper ions concentration were selected for fermentation conditions optima.Laccase properties were determined by studying the change of enzymatic activity.The result showed that the optimal temperature for laccase production was 25℃,copper ions concentration was 1.0 μmol / L.It was better to shaking the flasks with rate 120 r/min during fermentation.The optimum temperature for laccase activity were 30℃.Optimum pH value was varied with different sbustrates.Various metal ions showed different effects on the activity.The activity was enhanced by Cu2+,Co2+,K+ and Ag+,and was strongly inhibited by Fe2+,Fe3+and Ca2+.It suggested that glucose and peptone could be used as carbon and nitrogen for laccase production by T.pubescens MB89.The fermentation process of the existence of copper can accelerate the emergence of laccase.Substrates,temperature,pH,metal ions could influence the activity of laccase.
