Abstract
Objective:The paper aimed to construct a new T vector based on plasmid pUC19 digested with XcmⅠby molecular cloning methods.Method:Two complementary oligonucleotide chains containing two XcmⅠsites were synthesized.After denaturation and renaturation,the adaptor was cloned into plasmid pUC19 between the HindⅢand EcoRⅠsites.Result:The new plasmid,pUC19M vector,was digested with XcmⅠto derive a T-vector.The carrying capacity of pUC19M-T vector was 1kb by experimental verification.Conclusion:The T vector can be used in the gene cloing experiment.
