Abstract

AIM:To observe the effect of different concentrations of glucose and insulin on the secretion of VEGF in Colon26 tumor cells.METHODS:Colon26 tumor cells were cultured in vitro.According to the concentrations of glucose and insulin in culture medium,cells were divided into 12 groups.After added to different culture medium,cells were cultured for 48 h.In all experiments TGF-β1 concentration in the supernatant was detected by ELISA.RESULTS:In B,C,D groups(including 5,10,25 mmol/L glucose in culture medium,respectively),compared with A(culture medium,control),there was no statistical difference about the secretion of VEGF(P0.05).In G group(including 125 mU/L insulin in culture medium),the level of VEGF was higher significantly than A,E and F groups(including 0,5,25 mU/L insulin in culture medium,P0.01).However,no differences between A,E and F groups were found.In K and L groups(including 5,25 mmol/L glucose and 125,125 mU/L insulin in culture medium),the levels of VEGF were higher significantly than A(P0.01).However,no differences between K and L groups were found.In H,M and N groups(including 5,5,1.25 mU/L insulin and 5,25,25 mmol/L glucose in culture medium),compared with A,there was no statistical difference about the secretion of VEGF(P0.05).CONCLUSION:The concentration of glucose have no effect on secretion of VEGF from Colon26 tumor cells.High concentration of insulin may increase secretion of VEGF from Colon26 tumor cells.
