Microarray analysis on the differentially expressed genes in peripheral leucocytes from preeclampsia

Abstract

Objective:To analyze the differential gene expression profiles of peripheral blood leucocytes between preeclampsia and normal pregnancy cases using microarray technique.Methods:Total RNA in peripheral blood leucocytes was extracted by Trizol,and was used to make probes.After hybridization and washing procedure,the results of hybridization were scanned using computer system.Results:195 differential expression genes were detected among total 21 000 genes.74 genes were up-regulated and 121 were down-regulated.163 functional genes were involved in cell metabolism,signaling transportation,transcriptional control,cell proliferation and apoptosis.There were 20 unclassified genes and 12 unknown expressed sequence tag fragments.Conclusion:cDNA microarray technique is effective in screening out the differentially expressed genes between two different cases.Further analysis of those obtained genes will be helpful to understand the molecular mechanism of preeclampsia.
