Abstract
Objective:To construct the recombinant retroviral vector encoding anti-HCC single-chain bifunctional antibody gene (sFv-TNF-α)and establish a stable virus producing cell line.Methods:The sFv-TNF-α gene was cloned into a retroviral vector pLXSN and the recombinant vector was then transfected into the PA317 amphotropic packaging cells by calcium phosphate method.Results:The recombinant retroviral vector was constructed and a stable virus producing packaging cell line C 22 with high cfu(1×10 9·L -1 )was established.Conclusion:The packaged recombinant retrovirus can effectively transduce the targeted cells to transfer and express the exogenous genes.
