The Application of Chitosan in the Experimental Study for Bridging of Peripheral Nerve Defects by Nerve Guide

Abstract

To study the application of chitosan as tubing chambers(nerve guide) in nerve regeneration to bridge the sciatic nerve defects of rats.Ninety Wistar rats(200±20g)were chosen randomly,A 15mm defect was produced in the right sciatic nerve of each rat.Chitosan-NGF(A group),chitosan(B group) nerve guides were used to bridge the nerve defects,C group was the control group.The left side sciatic nerves were set up as the normal control group.Gross observation,microanatomical observation,light-microscopic and electro-microscopic histological examination,immunohistochemical examination and nerve electrophysiological examination were carried out at the end of 4th,12th, 24th weeks postoperatively.The results of nerve regeneration,vasculization,nerve fibers arrangement,myelination,nerve function reconstruction in A,B group were better than those in C group.The regenerated sciatic nerves of rats are provided with good regeneration microcircumstance by chitosan nerve guide.
