Application of Different Antigen Retrieval Methods in Immunohistochemistry Staining

Abstract

Objective:To select most appropriate antigen repairing method.Methods:In immunohistochemistry staining,5 monoclonal antibodies of mouse anti-human were selected with different antigen retrieval methods to determine the suitable antigen retrieval according to expressing situation of positive granules for different exposure dose,normal control,brain cortex and hippocampus exposure and paraffin tissue sections.Results:Antigen retrieval method affected directly the exposure of tissue antigen and masked or crosslinking modified antigen can be exposed or repaired by high pressure heating method to make immunohistochemistry results more accurate and reliable,especially nuclear positive tissue that was suitable for high pressure heating method.Positive result was poor with the electric boiling method and there was no difference between the positive results of enzymatic digesting antigen retrieval method and that of highly pressured heating antigen repairing method.Conclusion:Nuclear positive expression antibody should use highly pressured heating antigen repairing method.It is putative that the nuclear positive expression antibody should be also applied to the enzymatic digestion antigen repairing method.
