Abstract

Aim To investigate the effect of 2, 3-dioxoindoline on apoptosis and human telomerase reverse transcriptase mRNA in SH-SY5Y cells. Methods Change of nucleolus stained with Hochest 33258 was observed under fluorescent microscope;DNA fragmentwas observed with agarose gel electrophoresis; the expression levels of bcl-2 and bax protein were analyzed by Western blotting; the levels of hTERT mRNA ,bcl-2 mRNA and bax mRNA were examined by RT-PCR. Results After cells were treated with ISA of different concentrations (0,100,200,400 μmol·L-1) for 48 h, Hochest 33258 staining demonstrated apoptotic and normal cells. DNA ladder was found in cells treated with 200 μmol·L-1 and 400 μmol·L-1 ISA by agarose gel electrophoresis. The expressions of bcl-2 protein and mRNA were decreased as compared with that of the control (P0.05). However, the expressions of bax protein and mRNA kept constant (P0.05); and the expression level of hTERT mRNA , the key subunit of telomerase , decreased after 2, 3-dioxoindoline treatment compared with that of the control (P0.05). Conclusion 2, 3-dioxoindoline induces apoptosis of SH-SY5Y cells in a dose-dependent manner, which possibly results from the decreased expression of bcl-2 and hTERT.
