Abstract
Objective To investigate the effect of RelA antisense oligonucleotides (AsODN) on pancreatic carcinoma cell line Capan-2 proliferation. Methods The inhibition of pancreatic carcinoma cell proliferation was estimated by MTT and colony assay. RelA mRNA and RelA protein expression was detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry SABC method. The cell cycle was analyzed by flow cytometry (FCM). Results The cell growth inhibitory rate was 43.5 % afetr Capan-2 cell was treated with RelA AsODN for 48 h. The number of cells colony was reduced ( 47.3 ± 8.2 vs 26.7 ± 5.5, P 0.05 ). The expression of RelA mRNA and RelA protein (70.8 ± 6.5 vs 49.5 ± 6.0, P 0.01 ) was decreased significantly in Capan-2 cells with RelA AsODN. The number of G_0/G_1 phase cell was increased from 56.54 % to 82.83 %, and S phase cell reduced from 39.06 % to 11.29 %, Conclusion RelA antisense oligonucleotides exerts an inhibitory effect on pancreatic carcinoma cell proliferation.
