Abstract
Objective To explore whether the effects of high glucose on c fos,c Jun nucleoprotein levels and nuclear mitogen activated protein kinase (MAPK) activity were mediated by protein kinase c (PKC) activation in cultured rat glomerular mesangial cells.Methods Rat glomerular mesangial cells were cultured.The in situ PKC activity,c fos,c Jun nucleoprotein levels and nuclear MAPK activity were assayed after high glucose or high glucose with d α tocopherol for 24h,48h and 72h.Results Increased in situ PKC activity,c fos and c Jun nucleoprotein levels and nuclear MAPK activity were observed after 48h exposed to bigh glucose.All of them remained in the increased levels significantly after 72h except c fos nucleoprotein.All of the above mentioned effects of high glucose were inhibited by d α tocopherol.Conclusion High glucose may increase nucleoprotein levels of c fos、c Jun and nuclear MAPK activity mediated by PKC activation in cultured glomerular mesangial cells.D α tocopherol may ameliorated abnomal nuclear translocation signal induced by bigh glucose.
