Abstract
Objective:To investigate the effects of lysophosphatidic acid(LPA) on the proliferation and collagen synthesis of cardiac fibroblasts(CF) in vitro.Methods:Isolated and cultured neonatal Sprague-Daeley(SD) rats were used as experimental model.CF proliferation was measured by MTT assay.Collagen synthesis was measured by hydroxyproline alkaline hydrolysis assay.Results:①A_(490) value of CF by MTT assay increased with the elevation of LPA concentration.48 h later,only A_(490) value of 10~(-6) μmol/L group was significantly higher than that of control group(P0.05).72 h later,A_(490) value of 10~(-8),10~(-7),10~(-6) μmol/L groups were significantly higher than that of control group(P(0.05).) 96 h later,A_(490) value of 10~(-8),10~(-7),10~(-6) μmol/L groups were significantly higher than that of control group too.②The secretion of collagen increased with the elevation of LPA concentration and action time.48 h later,Only the collagen concentration of 10~(-6) μmol/L group was increased significantly compared to the control group(P(0.01).) 72 h later,the concentration of 10~(-6),10~(-7)μmol/L groups were increased significantly compared to the control group(P0.05,P0.01).96 h later,the concentration of all 3 treatment groups were increased significantly compared to the control group(P(0.05,) P0.01),the concentration of 10~(-6)μmol/L groups was increased significantly compared to that of the 10~(-7) and 10~(-8)μmol/L groups((P0.05)). Conclusion:LPA can increase the proliferation and collagen synthesis in rats.
