Inhibition of HeLa cell proliferation by cyclinE RNA interference

Abstract

Objective To study the effect of RNAi targeting cyclin E on the proliferation and cell cycle of cervical cancer cell line HeLa. Methods The eukaryotic expression vector of cyclin E RNAi was constructed and transfected into HeLa cell. The mRNA level of cyclinE was detected by RT-PCR. Cell proliferation was analyzed by MTT;Cell cycle was analyzed by flow cytometry. Results The vector of cyclinE RNAi was successfully constructed, and specially knocked down the mRNA level of cyclin E gene after transfection. MTT showed the cell proliferation was inhibited. Cells stopped at G0-G1 phase and cell number in S phase was sharply decreased after RNAi. Conclusion The mRNA level of cyclinE gene in HeLa cell can be specially knocked down by RNAi, and the malignant proliferation of cervical cancer cells is inhibited too.
