Effect of cyclosporin on change of podocyte motility stimulated by lipopolysaccharide

Abstract

AIM To observe the protective effect of cyclosporin(CsA) on the change of podocyte motility stimulated by lipopolysaccharides(LPS).METHODS Cultured podocytes were divided into 3 groups.Control group:the podocytes were cultured without any treatments;LPS group:the differentiated podocytes were cultured with LPS(50 mg·L-1) for 24 hours;LPS + CsA group:the podocytes were cultured with LPS(50 mg·L-1) and CsA(0.5 mg·L-1) for 24 hours.The change of podocytes motility was detected by immunofluorence method,flow cytometry method,transwell migration assay and wound healing assay,etc.RESULTS There was no significant difference among the three groups in synthesis of the integrin beta-3.The expression of AP5 and the average number of podocytes were((1.25 ± s 0.12)%,(177 ± 7)) in the control group,((52.47 ± 0.12)%,(328 ± 24)) in the LPS group,and((10.28 ± 0.28)%,(232 ± 10)) in the LPS +CsA group,respectively.There were significant differences among the three groups(P 0.01).Compared with the control group,the activation of integrin beta-3 was higher in the LPS + CsA group(P 0.01),but there was no significant difference in the migration between the two groups(P 0.05).CONCLUSION In the normal condition,differentiated podocytes are in a quiescent state.When podocytes are stimulated by LPS,integrin beta-3 is activated with the motility increasing and the adhesion decreasing.CsA could stabilize podocytes through decreasing the activation of integrin beta-3 and reducing the ability of podocytes motility.
