Abstract
Objective To explore the biological characteristics of mouse adipose tissue-derived mesenchymal stem cells(ADMSCs) from the inguinal subcutaneous adipose tissues and paraepididymal adipose tissues.Methods ADMSCs were isolated from the inguinal subcutaneous adipose tissue and the paraepididymal adipose tissue and then cultured in vitro.Cellular morphology was observed;cell harvest was calculated;cell growth was measured by MTT assay;and the cell surface marker CD44 was detected by flow cytometry.After the cells were induced by dexamethasone and insulin,the intracellular neutral lipids were detected with oil Red O staining.Results Most of the ADMSCs were of spindle shape.The amount of ADMSCs differed between the two different tissue sources.The rate of cell adhesion was similar at different time points.ADMSCs from different tissue sources had the similar capability to proliferate.A majority of the cells arrested in the G0/G1 phase.CD44 was positive in all of the ADMSCs.After adipogenic induction,lipid droplets were formed in the cytoplasm,with bright red color for oil Red O staining.Conclusions The amount of ADMSCs from the paraepididymal adipose tissue was greater than that from the inguinal subcutaneous adipose tissue per unit mass.But other biological characteristics of the ADMSCs and the ability of the cells to differentiate into the adipogenic lineage do not differ significantly.
