Abstract
OBJECTIVE To study the physicochemical characteristics and hemostatic effect of the self-producing water-soluble O-carboxymethyl chitosan hemostatic sponge.METHODS According to the prescription,O-carboxymethyl chitosan hemostatic sponge was prepared by using the vacuum freeze-drying technology.The physicochemical characteristics of the self producing sponge were observed by outward appearance,internal aperture,porosity,water-absorptivity,the density and so on.With establishing a wounding model by femoral artery shearing of the rats,commercial absorbable-gelatin sponge was used as a positive control,and medical gauze was used as a negative control.The efficacy of hemostasis was compared between the self-producing sponge group and the controls;their hemostatic reliability was observed.RESULTS The self-producing sponge outward appearance was loose texture,porous,off-white color and good toughness,the internal aperture was probably 100-200 μm,its porosity achieved above 65.11% and the water-absorptivity was probably 38.29%,the density was 0.045 g·cm-3.The bleeding time of the self-producing sponge was significantly shorter than that of the commercial absorbable-gelatin sponge(P0.01),moreover the recurring hemorrhage of the self-producing sponge was fewer in the rats with femoral artery shearing.CONCLUSION The results showed that the self-producing O-carboxymethyl chitosan hemostatic sponge has good physicochemical characteristics,its hemostatic effect is remarkable,and it can be used in clinical practice.
