Abstract

Objective To investigate the effect and significance of cardiotrophin-1(CT-1) in the hypertrophy and lesion of myocardial cells induced by angiotensinⅡ(AngⅡ). Methods The cultured myocardial cells were treated with AngⅡ to induce hypertrophy. The morphologic changes of myocardial cells were observed under contrast phase microscope and electron microscope. The mRNA levels of CT-1 and β-MHC were examined by RT-PCR method. The protein expressions of CT-1 were examined by immunohistochemistry method. The apoptosis of myocardial cell was identified by ELISA. Results After stimulated by AngⅡ, the myocardial cells enlarged. Both the mRNA levels of β-MHC and the apoptosis increased. The mRNA level and protein expression of CT-1 increased. AG490 may partially inhibited the myocardial cell hypertrophy and decreased the apoptosis related with CT-1 Conclusions CT-1 may take part in the hypertrophy of myocardial cells. AG490 inhibits the hypertrophy and does not decrease the protective effect of CT-1. [
