Abstract
Objective To investigate the effect of Human telomerase reverse transcriptase (hTERT) gene antisense phosphorothioate oligodeoxynucletide (ASODN) on telomerase activity in K562 cells. Methods By using liposome mediated DNA transfection, telomerase antisense RNA was successfully introduced into k562 cells. The telomerase activity of K562 cells was examined by telomeric repeat amplification protocol(TRAP)-enzyme-linked immunosorbent assay(ELISA).The expression levels of hTERT mRNA protein was assayed by immunofluorescence using fluorescence isothiocyanate label. Results TRAP- ELISA and immunofluorescence assay showed that the telomerase activity and expression levels of hTERT mRNA protein from K562 cells decreased with time after hTERT ASODN treatment. Conclusion It seems to be a new method to treat leukemia by using telomerase antisense to inhibit telomerase activity of K562 cells, telomerase is a new latent approach to treat leukemia.
