Effects of carvedilol on preventing left ventricular remodeling in chronic heart failure of juvenile rats

Abstract

Objective To compare the effect of carvedilol in large,middle,or small dose in preventing left ventricular remodeling in chronic heart failure of juvenile rats.Method The animal model of chronic heart failure was established by constriction of abdominal aorta.The 36 surviving malerats of five weeks old were randomly divided into 4 groups:(1)CHF control( n =8);(2)large dose carvedilol (10mg·kg -1 ·d -1 , n =10);(3)middle dose carvedilol (1mg·kg -1 ·d -1 , n =10);(4)small dose carvedilol(0.1mg·kg -1 ·d -1 , n =8).Sham operated rats( n =8) were selected randomly as non infarction control.Carvedilol was administered bydirect gastric gavage.The weight of rats was observed every week.After 4 weeks of drug therapy,the highfrequency ultrasonography(HFUS),and the hemodynamic studies were performed,and the hearts of the juvenile rats were taken out in order for pathologic analysis.Results Compared with the sham operated group,LVESD,IVSTd,IVSTs,LVPWTd,LVPWTs,LVRW,RVRW,SBP,DBP,MBP,LVSP,LVEDP all sinificantly increased (all, P 0.01) in CHF group,as well as LVEDD( P 0.05 ).A significant decrease could be seen in Weight,FS,+dp/dt max ,-dp/dt max .In comparsion with CHF groups,a dose dependent and significant decrease could be seen in LVEDD,LVESD,IVSTd,IVSTs,LVPWTd,LVPWTs,LVRW,RVRW,SBP,DBP,MBP,LVSP and LVEDP,especial large and middle dose carvedilol groups(all P 0.05~0.01).While a significant increase could be seen in Weight,EF,FS,+dp/dt max ,-dp/dt max .( P 0.01).Conclusion Carvedilol in no matter what dose effectively and dose dependently prevent LV remodeling in chronic heart failure of jurvenile rats and hemodynamics LV funnction,the weight of body in rats.
