EFFECTS OF HUJIN DECOCTION ON MMP-2mRNA AND TIMP-2mRNA EXPRESSION IN LUNGS OF RATS WITH ACUTE LUNG INJURY

Objective:To discuss the effects of hujin decoction on expression of metalloproteinase-2mRNA(MMP-2mRNA)and tissue inhibitor of metalloproteinase-2mRNA(TIMP-2mRNA)of lungs of rats with acute lung injury caused by LPS.Methods:40 rats were divided into 4 groups,normal,model,dexamethasone and hujin decoction groups,each group was divided into two subgroups after LPS-induced 4 and 8 hours,dexamethasone and hujin decoction groups were respectively given dexamethasone and hujin decoction before injecting LPS to induce acute lung injury.The lung's index was determined,the expression of MMP-2mRNA and TIMP-2mRNA of lungs were examined by real-time fluorescent PCR method,the histopathology changes of lungs were observed by light scope.Results:To compare with model group,hujin decoction could lowered the lung's index of 4 and 8 hours groups,the expression of MMP-2 mRNA of hujin decoction group was lowered,too,the expression of TIMP-2mRNA was raised obviously.There was large piece of pulmonary hemorrhage and necrosis was seen in the model group,hujin decoction could improve the pathological changes.
