Effects of acetazolamide on proliferation, IL -1β expression, and TNF -α expression of fibroblast-like synoviocytes of rheumatoid arthritis

Abstract
To observe the effect of acetazolamide on proliferation,IL-1β expression,and TNF-α expression of fibroblast-like synoviocyte(FLS) of rheumatoid arthritis(RA),FLS were separated from synovia and cultured in vitro.Then,RA FLS was treated with different concentrations of acetazolamide(10-4,10-6,10-8 mol/L) for different times(24,48,72 h).We employed MTT method to study the growth inhibition of RA FLS,ELISA to detect the concentration of TNF-α and IL-1β in cell-cultured supernatant fluid.Our results showed there was no significant difference in absorbance value of RA FLS treated with different concentrations of acetazolamide(P 0.05),but all concentrations of acetazolamide could inhibit the proliferation of RA FLS in a time-dependent manner(P 0.05).Furthermore,different concentrations of acetazolamide could significantly inhibit the expression levels of IL-1β and TNF-α in cell-cultured supernatant fluid of RA FLS(P 0.05) in a time-dependent and dose-dependent manner.In conclusion,acetazolamide could inhibit the proliferation of RA FLS and decreased the secretion of TNF-α and IL-1β in cell-cultured supernatant fluid of RA FLS.
