Abstract
Objective:To observe mRNA and protein expression of Msx2 in kidney tissues of the rats with glucocorticoid-induced osteoporosis(GIOP)and discuss the pathogenesis of GIOP and the therapeutic effect and mechanism of reinforcing the kidney to replenish marrow traditional Chinese herbs compound.Methods:We established the GIOP rat models by intramuscular injection of dexamethasone.We divided all the rats into normal group,blank model group,reinforcing the kidney to replenish marrow traditional Chinese herbs compound group,reinforcing the middle to benefit qi granula group,removing blood-house stasis capsule group and Gushukang granula positive control group.After 9 weeks of establishing models and administrating by gavage,we detected femur bone mineral density by using the XR-26 dual energy X-ray absorptiometry.We detected mRNA expression of Msx2 in kidney tissues by Real-Time Quantitative RT-PCR and detected their protein expression by Western Blot.Results:Compared with normal group,femur bone mineral density of blank model group decreased significantly(P0.01),mRNA and protein expression of Msx2 decreased significantly(P0.01)in kidney tissues;Compared with blank model group,femur bone mineral density of reinforcing the kidney to replenish marrow traditional Chinese herbs compound group increased significantly(P0.01),mRNA and protein expression of Msx2 up-regulated significantly(P0.01)in kidney tissues.Conclusions:Maybe it is one aspect of the pathogenesis of GIOP that mRNA and protein expression of Msx2 decreases in kidney tissues;The reinforcing the kidney to replenish marrow traditional Chinese herbs compound can prevent and treat GIOP effectively by up-regulating mRNA and protein expression of Msx2 in kidney tissues.
