Experimental study on the injury of human umbilical vascular endothelial cells induced by high glucose

Abstract

Objective To study the effects of constant and intermittent high glucose on the proliferation and apoptosis of human umbilical vascular endothelial cells( HUVECs). Methods HUVECs were divided into three groups: the control group( 5. 5 mmol·L-1),constant high glucose group( 30. 5 mmol·L-1),intermittent high glucose group( every 24 h rotation by adding 5. 5 mmol · L-1or 30. 5 mmol · L-1glucose solution). The cell proliferation and apoptosis was detected respectively by CCK-8 assay and flow cytometry,and the cell morphology was observed by inverted phase contrast microscope. Results Constant high glucose and intermittent high glucose could inhibit the cell proliferation and induce its apoptosis,as compared with the control group( P 0. 05). The viability of HUVECs induced by the intermittent high glucose is lower than that of the constant high glucose,but the apoptosis is higher. Conclusion High glucose could inhibit the proliferation of HUVECs and promote the apoptosis. Compared with the constant high glucose,intermittent high glucose could be more deleterious to HUVECs.
