Determination of Eugenol,Costunolide and Dehydrocostuslactone in Qiwei Guangzao Pills by HPLC

Abstract

OBJECTIVE To establish a HPLC quantitative method for the determination of eugenol,costunolide and dehydrocostuslactone in Qiwei Guangzao pills.METHODS The separation was performed on a C18 column(Agilent ZORBAX Eclipse SB-C18,4.6 mm× 150 mm,5 μm),with the mobile phase consisted of 5% acetonitrile-acetonitrile(58∶42).The flow rate of 1 mL·min-1.The detective wavelength was set at 280 nm(0~15 min),225 nm(15.1-40 min).RESULTS The retention times of eugenol,costunolide and dehydrocostuslactone were about 6,21 and 24 min.The resolution was above 1.5.The regress equation for eugenol was Y=0.104 5ρ-6.161,r=0.999 9,and the linear range was 0.552 0-13.80 μg.The equation of costunolide was Y=0.095 39ρ-0.297 2,r=0.999 9,and the linear range was 0.074 00-1.850 μg·mL-1.The equation of dehydrocostuslactone was Y=0.078 51ρ-0.079 49,r=0.999 8,and the linear range was 0.059 50-1.488 μg.The average recoveries of eugenol,costunolide and dehydrocostuslactone were 99.2%,101.3% and 97.0% with RSDs of 0.89%,1.6% and 1.6%.CONCLUSION This method is simple,time-saving and accurate.It can be used for routine analysis of eugenol,costunolide and dehydrocostuslactone in Qiwei Guangzao pills.
