Expression and significance of HSP70,IL-6 and TNF-α in the tissue of acute liver enjury induced by CCl_4

Abstract

Objective To investigate the action of HSP70,IL-6 and TNF-α in the acute liver injury and regeneration.Methods One hundred mice were randomly divided into the observed group(n=90)and control group(n=10).In the observed group,acute liver injury of mice were induced by abdominal injection of CCl_4,10 mice were executed at 3,6,12,24,30,36,42,48,54 h after CCl_4 injection.The serum ALT and AST levels were determined;the pathological changes of liver tissue were observed by HE staining method;the protein expression of HSP70,IL-6,TNF-α were detected by Western blotting.Results Compared with the control group,the activity of ALT and AST in the observed group were significantly increased,peaked at 36 h,then decreased gradually to the normal level.Compared with the control group,the protein expression of HSP70,IL-6 and TNF-α were significantly changed in the observed group: the protein expression of HSP70 were significantly increased,peaked at 12 h,reduced after 24 h,then increased after 36 h;the protein expression of IL-6 were significantly decreased,increased after 24 h,peaked at 30 h,then decreased gradually,but higher than the normal level at 54 h;the protein expression of TNF-α were significantly increased,peaked at 30 h,reduced after 36 h,but much higher than the normal level.Conclusions The protein expression of HSP70,IL-6 and TNF-α significantly changed during the process of acute liver injury and regeneration,these factors probably play important roles in the occurrence and development of acute liver injury.
