Abstract
Aim To study the effects of total flavonoids of scutellaria baicalensis georgi(TFSB)on bleomycin-induced pulmonary fibrosis in rats and investigate its mechanisms.Methods The pulmonary fibrosis model was estalished by intratracheal injection of Bleomycin.After 28 d,rats were treated with TFSB by intragastric administration,and the pathomorphology,the biochemistrical index in lung tissue and blood were measured and analyzed.The effects of TFSB on the expressions of transforming growth factor-β1(TGF-β1),Smad2/3 mRNA and the expressions of alpha-smooth muscle actin(α-SMA)and peroxisome proliferator-activated receptor-gamma(PPAR-γ)protein in lung tissue of rats were assayed by RT-PCR and Western blot.Results Compared with the model group,the contents of MDA in serum and lung tissue and the contents of HYP were decreased(P0.05,P0.01),the contents of GSH,T-AOC and SOD in serum were increased(P0.05,P0.01),the contents of GSH in lung tissue were increased(P0.01).TFSB could obviously reduce alveolitis and pulmonary fibrosis(P0.05,P0.01).The expressions of TGF-β1 and Smad2/3 mRNA in lung tissue of rats were decreased(P0.05),and the expression of α-SMA in lung tissue of rats was decreased(P0.05).In contrast,the expression of PPAR-γ protein in lung tissue of rats were increased(P0.05).Conclusion TFSB has therapeutic effects of bleomycin-induced pulmonary fibrosis in rats,which might be related with the anti-oxidation and the inhibition of TGF-β1 signaling pathway.
