Abstract

Objective To study the method of isolated culture of bone marrow mesenchymal stem cells(MSCs),to establish the rat model of hepatic ischemia-reperfusion,and observe the effect of MSCs to the hepatic ischemia-reperfusion injury.Methods MSCs of rats were cultured in vitro,rats were divided randomly into the treated group and the negative control group.The treated group were injected MSCs 0.5 ml,the control group were injected normal saline 0.5 ml.The normal rats were blank control group.Results All animals blood were collected after 1 and 2、4weeks to observe the serum activities of ALT,AST,MDA and SOD.At first week,the serum activities of ALT、AST and MDA in treated group and negative control group were higher than the blank control group(P0.05).There's no much difference between the treated group and negative control group(P0.05).At second week,the serum activities of ALT,AST and MDA in treated group were lower than negative control group(P0.05),but the serum activities of SOD in treated group was higher than negative control group(P0.05).At four week,the serum activities of ALT,AST and MDA in treated group were lower than negative control group(P0.05),and ALT,AST have no difference between blank control group(P0.05),but MDA,SOD have much difference(P0.05).Conclusion MSCs can promote cells proliferation of the hepatic ischemia reperfusion injury.
