Abstract
Objective: To investigate the effect of CpG motif oligodeoxynucleotides (CpG ODN) on the antigen induced allergic airway reaction in mice. Methods: The asthma model was set up in the C57BL/6 mice with OVA, the CpG ODN in the dose of 30 μg was co administered intraperitoneally with the antigen in sensitization stage to study its effect on the airway allergenic reactions. Results: (1)Compared with the control, coadministration of CpG ODN in sensitization phase significantly inhibited airway eosinophilia after antigen challenge( P 0.01). (2) Coadministration of CpG ODN with antigen reduced the concentrations of IL 4, IL 5 in the bronchoalveolar lavage accompanied with increase of IFN γ and IL 12 after antigen challenge.(3)CpG ODN with antigen prevented the development of bronchial hyperreactivity. (4)Coadministration of CpG ODN also reduced the total and antigen specific IgE levels in serum( P 0.05). Conclusion: This study suggests that intraperitoneal coadministration of CpG ODN with antigen during sensitization stage can inhibit antigen induced airway allergic reactions significantly, preventing the development of asthma by involving increase of production of IL 12 and IFN γ induced by CpG ODN.
