Abstract
Objective To study the effects of the level of chimerism and relation between donor and recipient HLA match on graft-versus-host disease after hematopoietic stem cell transplantation.Methods BALB/C mice were used as the recipient.BALB/C mice were pretreated 3 days before transplantation with cidomycin.Group A was pretreated with TBI(4.0 Gy),and group B was pretreated with TBI(8.0 Gy).The donor of both group were CB6F1 mice.BALB/C mice pretreated with TBI(8.0 Gy) were the recipient.CB6F1 mice(Group C) and C57BL/6 mice(Group D) were donors.And bone marrow cells were injected through all BALB/C mice tail vein,and the prevention of GVHD was not given.For all recipient mice,the levels of Chimerism and GVHD were observed.Results The results of chimera showed that engraftments of group A remained mixed donor chimerism and unstable,tended to decrease after 14 days.Engraftments of group B,C and D were full donor chimerism,and the chimerism of group B,C and D remained above 90 % and preserved even after 35 days.The clinical GVHD score in group B was obviously higher than that of group A(P0.05),and those in group D higher than group C(P0.05).Conclusions GVHD has a association with the level of Chimerism and relation between donor and recipient HLA match after hematopoietic stem cell transplantation.
