Influence of Shenluotong serum on secretion of ECM and expression of CTGF mRNA of mesangial cells induced by angiotensin in vitro

Abstract

Objective To investigate the inhibitory effect of Shenluotong serum on the different compositions of extracellular matrix(ECM) and the influence on the connective tissue growth factor(CTGF) in mesangial cells(MCs).Methods The contents of fibronectin(FN) and collagen Ⅳ(Col Ⅳ) in ECM were detected by enzyme linked immunosorbent assay(ELAIS),and the expression of CTGF mRNA was measured by semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) based on cultured in vitro rat glomerular mesangial cells and the method of serum pharmacology.Results The contents of FN、Col Ⅳ in ECM and expression of CTGF mRNA were showed positive correlation and all inhibited by Shenluotong significantly.Conclusion Shenluotong can diminish the accumulation of ECM,and inhibit the secretion and expression of CTGF,which maybe one of the mechanisms of preventing the kidney from sclerosis.
