Abstract
Objective To investigate the expression of Toll-like receptors (TLR 3, TLR7, TLR8 and TLR9) mRNA in peripheral blood mononuclear cells (PBMC) from COPD patients.Methods PBMC from both COPD patients and healthy donors were obtained by density gradient centrifugation. After isolation of total RNA from the PBMC, cDNA was synthesized by reverse transcription of total RNA. Copy numbers of TLR3, TLR7, TLR8 and TLR9 were determined by real-time quantitative PCR.Results The expression of TLR3 and TLR9 mRNA in PBMC were significantly increased in COPD patients as compared with healthy donors(P0.05).There were no significant differences in expression of TLR7 and TLR8 mRNA in PBMC between COPD patients and healthy donors(P0.05). No significant differences were observed in expression of TLR3, TLR7, TLR8 and TLR9 mRNAs between acute phase COPD patients and stable phase COPD patients(P0.05).Conclusion Up-regulation of TLR3 and TLR9 in COPD patients might be an important mechanism for increased sensitivity to virus infection.
