Effects of VEGF ASODN delivered by Oligofectamine on the proliferation and apoptosis of PC3 cell line

Abstract

Objective To detect whether VEGF ASODN delivered by oligofectamine could affect the proliferation and apoptosis of human prostate cancer PC3 cell line.Methods VEGF ASODN was delivered into PC3 cells by oligofectamine.There were three groups in the experiment: VEGF ASODN,VEGF ODN and control.Quantitative RT-PCR and Western Blot assay were used to detect the VEGF expression.MTT assay and flow cytometry were used to test the cell proliferation and apoptosis.Results Compared to control group and ODN group,the cells of VEGF ASODN expressed lower VEGF mRNA and protein.Moreover,the cell proliferation was inhibited(P0.05) and apoptosis percentage increased(P0.05).Conclusion VEGF ASODN delivered by oligofectamine could inhibit VEGF expression of PC3 cells,which resulted in that the cell proliferation was inhibited and apoptosis increased.
