Abstract
Objective:To explore chemical hypoxia effect of cobalt chloride on endothelial cells injury of human umbilical vein and expression of soluble vascular endothelial growth factor receptor-1(sFlt-1) mRNA.Methods:Proliferation of endothelial cells of human umbilical vein was detected by MTT assay,lactic dehydrogenase(LDH) released from all endothelial cells of human umbilical vein into the medium were measured by LDH kits;the expression of sFlt-1 was detected by quantitative reverse transcriptase-polymerase chain reaction(RT-PCR).The relationship between hypoxia and proliferation of endothelial cells of human umbilical vein,the level of LDH,the expression of sFlt-1Results:Cobalt chloride could injury endothelial cells of human umbilical vein by inducing the expression of mRNA of sFlt-1,which depended on hypoxia time.Conclusion:The expression of sFlt-1 mRNA in injuried endothelial cells of human umbilical vein increases,which promotes the onset of hypertensive disorder complicating pregnancy.
