Abstract

Objective To study the effects of Arsenic trioxide(As2O3)on the human renal cell carcinoma A-704 cell line.Method A-704 cell line were cultured in vitro and were influenced by As2O3 in the different concentration. The cells were detected with microscope,DNA gel electrophoresis and MTT staining respectively to observe the anti-proliferative and apoptosis effects. Results As2O3 inhibited the growth of cells, showed the apoptotic change and DNA change in a concentration-dependent manner.Treated with As203for 96 h at the concentrations of 0.5,1.0,2.0,4.0 μmol/L,the inhibition rates of As203 on A-704 cells were 40.01%,43.72%,47.54%,53.61% respectively.Compared with untreated cells, there are magnificent differences at each concentration(P﹤0.05).Conclusion As203 significantly inhibited the proliferation of A-704 cells by inducing apoptosis in these cells.
