Effect of propofol on S100βprotein,total calcium content,and water content in the early stage of acute brain injury in rats

Abstract

Objective To observe the effect of propofol on S100βprotein,total calcium,and water content in the early stage of acute brain injury in rats.Methods Forty-eight healthy male Sprague-Dawley rats were randomly divided into six groups with 8equal rats each as follows:pseudoperation group(group S),brain injury group(group I),fat emulsion group(group F),and different concentrations of propofol groups(group L, M,and H,propofol 17.46,34.92,and 69.84 mg·kg-1·h-1 continuous infusion,respectively).Models of acute brain injury were made according to Feeneys methods.Serum S100βcontent at 15min(T1)and 60min(T2)after brain injury were measured,as well as total calcium and water contentin injured brain by decapitation 60min after the model established.Results Compared with group S,S100βprotein and total calcium content increased significantly in the other five groups at T1and T2(P0.05),besides,brain water content increased in groups I,F,and H(P0.05).S100βprotein decreased in groups L,M,and H at T2compared with that of T1(P0.05).Compared with group I,S100βprotein and water content decreased in groups L,M,and H(P0.05)as well the water content in group L(P0.05).Similarly,compared with group F,S100βprotein and water content decreased in groups L,M,and H as well the water content in group L(P0.05).However,compared with group L,S100βprotein increased at T1in group H and decreased at T2in groups M and H(P0.05),otherwise,the total calcium content decreased significantly in groups M and H(P0.05),and the water content increased in group H(P0.05). Further,compared with group M,S100βprotein increased at T1and T2in group H(P0.05),which was concomitant with a increased calcuium content and water content(P0.05).Conclusion Early application of propofol can reduce the serum S100βprotein contents,the total calcium content,as well as the water content,especially with a dose of 34.92mg·kg-1·h-1.
