Comparison of Two Extracting Methods for Soil Microbial Total DNA

Abstract

DNAs of soil microbes under rubber plantations were extracted by using two methods, CTAB-SDS extraction and SDS extraction, and the results of these two methods were compared. Using these two extraction methods a 23.1 kb fragment of DNA was extracted. There was a very high difference between these two methods in colour, DNA yields, DNA purity, etc. The first method had a better purity in extracting DNA which was directly used for PCR amplification and digestion of restriction enzymes and later operation without purification. The second one was low in the purity, and was not directly used for PCR amplification. These two methods are fit for DNA extraction of the microbes from the soil of rubber plantations. For purity of the total DNA of the soil microbes, the first method is the best choice. For the DNA extraction of the soil microbes under the rubber plantations, the first method is recommended.
