Abstract
AMP-activated protein kinase(AMPK) plays a central role in maintaining energy homeostasis in cell metabolism.For more than twenty years,researchers have used 32P-labeled SAMS peptide assays to assess AMPK activity in vitro.However,the isotope radiation is dangerous and can cause radioactive contamination of the environment.This paper reports the development of a convenient non-radioactive label for AMPK activity in vitro for daily lab use.The method was used to compare endogenous AMPK activities from different mouse tissues to show the endogenous AMPK activity varies greatly among tissues.The results show that this method can be used for further studies of AMPK enzyme activity and the interactions of proteins with AMPK.
