Abstract
A study on Agrobacterium tumefaciens mediated transformation of wheat was performed by using six Agrobacterium tumefaciens strains, different wheat genetypes, different explants and recombinant plasmids pCAMBIA3301, which contain gus and bar genes. Two highly infectious Agrobacterium tumefaciens strains to wheat and some highly sensitive wheat acceptors were obtained. After cocultivation and two steps selection on a media with herbicides wipeout(20 mg/L) and PPT(3.75 mg/L) respectively, 70 plantlets with PPT resistance were obtained. some of them proved transgenic plants by molecular analysis.
