An Animal Model of Collagenase-Induced intracerebral Hemorrhage in Mice

Abstract

Objective To establish a typical mouse model of intracerebral hemorrhage.Methods Fourty mice were randomly divided into normal group and model group.Intracerebral hemorrhage in mice were generated by stereotaxically injecting collagenase typeⅦ.Thirty adult mice,each received a 0.15 μL injection of heparinate-bacterial collagenase mixture(0.075 U collagenase typeⅦ and 0.318 U heparinate)into the right caudate nucleus.Neurological scores were assessed and recorded at 8,12,24,72 hours after injection.After behavioral testing the brains were processed for routine histological examination. The diameter of hemorrhage area on the coronal section via the injecting point was measured.Results After the mice wake up,they dipicted neurological impairments.The defects became more and more seriously in the first 24 hours after operation.Histological studies revealed that the hemorrhage peaked within 24 hours.Conclusions Intracerebral hemorrhage can be induced in mouse by injecting collagenase into the caudate nucleus of the brain.This is an useful economical and reproducible method to study intracerebral hemorrhage.Mouse is a valuable laboratory animal to be used in studies of intracerebral hemorrhage.
