Abstract
Objective: To study the immunosuppression and antitumor activity of FTY720 to hepatocellular transplanted tumor in mice. Methods: Mice liver cancer Hepa1-6 cells were collected and injected subcutaneously with a dose of 2 × 106/mouse into C57BL/6j mice. When the diameter of the hepatocellular transplanted tumor reached 0.30 cm, 40 C57BL/6j mice were randomly divided into four groups, each group was lavaged for 10 days with the following medicine separately, Ciclosporin group: Ciclosporin 10 mg/(kg·d); FTY720 group: FTY720 3 mg/(kg·d); 5-Fu group: 5-Fu 50 mg/(kg·d); NS group: NS 1 mg/(kg·d). Then thymus gland and spleen index, the percentage of peripheral blood T-lymphocyte, gross tumor volume, secretory volume of AFP in serum, apoptosis index as well as the expression of Caspase-3 and Bcl-2 were investi gated. Results: Thymus gland and spleen index and the percentage of T-lymphocyte in Ciclosporin group and FTY720 group were lower than that of NS group (P0.05, P0.01), there was no significant difference between NS group and 5-Fu group (P0.05); the gross tumor volume and secretory volume of AFP in serum in 5-Fu group and FTY720 group were lower than that of NS group (P0.05, P0.01), and the apoptosis index was higher than that of NS group (P0.05, P0.01), there was no significant difference between NS group and Ciclosporin group (P0.05); the expression of Bcl-2 in 5-Fu group and FTY720 group was lower than that of NS group (P0.05), there was no significant difference between NS group and Ciclosporin group (P 0.05); the expression of Caspase-3 in 5-Fu group was lower than that of NS group (P0.01), there was no significant difference between NS group and FTY720 group as well as Ciclosporin group (P0.05). Conclusion: FTY720 has immunosuppression and antitumor activity, the antitumor activity is associated with the lower expression of Bcl-2.
