Expression of MCP-1,TGF-β_1 and CTGF in the Renal Tissues of Rats with Type 2 Diabetic Nephropathy

Abstract
Objective To investigate the expression of monocyte chemotactic protein-1(MCP-1),transforming growth factor beta1(TGF-β1) and connective tissue growth factor(CTGF) in the renal tissues of rats with STZ induced type 2 diabetic nephropathy.Methods Thirty-five Sprague-Dawley male rats were randomly divided into control group(N group,14 cases) and diabetic nephropathy model group(DN group,21 cases).The DN group include rats with insulin resistance induced by diets,the rats were given a single intraperitoneal injection of streptozotocin of STZ.The rats in N group were given conventional feed,and were given a single intraperitoneal injection of the same amount of citric acid-sodium citrate buffer.Immunohistochemisty were performed to examine the expression levels of three factor in renal tissue.Results The expression levels of MCP-1,TGF-β1 and CTGF were strong in the renal tissues of rats with STZ induced type 2 diabetic nephropathy,the average absorbance of three factors in the DN group was(0.29 ± 0.04),(0.20 ± 0.02),(0.20 ± 0.02) respectively,the average absorbance in the N group was(0.21 ± 0.02),(0.17 ± 0.01),(0.16 ± 0.02) respectively,there were significant difference between DN group and N group of three factors(P 0.05).Conclusion The expression of MCP-1,TGF-β1 and CTGF were obviously increased in the renal tissues of rats with STZ induced type 2 diabetic nephropathy,which are involved in the pathogenesis of early diabetic nephropathy.
