AEPS induces apoptosis in SMMC-7721 hepatocellular cancer cells and its mechanism

Abstract

Objective:To study the mechanism of apoptosis induced by AEPS in SMMC-7721 hepatocellular cancer cells.Methods:The fluorescent microscope was utilized to observe the morphological changes of apoptosis,apoptosis rate were detected with flow cytometry.Western blot was utilized to detect the expression of survivin,caspase-3 and bcl-2.Results:Typical morphological changes of apoptosis in SMMC-7721 cancer cells could be observed by the fluorescence microscope.Compared with that of the control cells,apoptotic rate of the cells treated with AEPS was increased signifcantly(P0.01)and in a dose-and time-dependent manner.Western blot showed that survivin and bcl-2 expression was signifcantly decreased(P0.01),whereas caspase-3 expression was signifcantly increased(P0.01)in SMMC-7721 cancer cells treated with AEPS.Conclusion:AEPS can markedly induce apoptosis in SMMC-7721 hepatocellular cancer cells.The mechanism might be related to the change of survivin,caspase-3 and bcl-2 expression.
