Abstract

AIM:To observe the effects of P.gingivalis and P.intermedia LPS on the production of IL-6 and IL-8 from human gingival fibroblast cells and the effect of LPS on hGFs was mediated by mCD 14 or not. METHODS: Human gingival fibroblasts were stimulated with Pg/Pi -LPS of different concentrations and time periods. The activity of IL-6 and IL-8 in the culture medium were quantitated by ELISA. In addition, RT-PCR has been used to identify the expression of mRNA of IL-6, IL-8, mCD 14 in hGFs. RESULTS: Under the stimulation of LPS of P.gingivalis and P.intermedia significant greater amounts of IL-6 and IL-8 were released in a dose and time dependent way than those of the unstimulated cells. The levels of IL-6 and IL-8 mRNA were also increased, but CD 14 mRNA message was undetectable. CONCULSION: LPS from periodontopathic bacteria, P.gingivalis and P.intermedia , was capable of stimulating hGF cells to release cytokines.Furthermore,the LPS effect on hGFs was not mediated by CD 14 .
