Determination of Calycosin-7-Glucoside in Astragali Radix from Different Place by HPLC

Abstract

Objective To determine Calycosin-7-Glucoside in Astragali Radix from different place by HPLC so as to provide experimental basis for quality evaluation of Astragali Radix.Methods HPLC was used in the quantitative analysis of Calycosin-7-Glucoside.The measurement conditions were as follows: Diamonsil C18(250 mm × 4.6 mm,5 μm) column; mobile phase: acetonitrile-0.2% formic acid aqueous solution by gradient elution; fl ow rate: 1.0 mL·min-1; detection wavelength: 260 nm,column temperature: 30 ℃.Results There was a good linear relationship between the concentration of Calycosin-7-Glucoside and absorption area value in range of 9.92 μg·mL-1~ 248.00 μg·mL-1(r =0.999 5).The average recovery was 97.70% with RSD=2.64%.Conclusion There was great difference in Calycosin-7-Glucoside content of Astragali Radix from different place.HPLC method is simple and accurate,and can be widely used in the quality evaluation of Astragali Radix.
