Abstract

Objective To investigate therapeutic effects and mechanisms of Hydrocortisone on severe acute pancreatitis(SAP).Methods Fifty-four SD rats were randomized into three groups:the sham operation(SO) group, SAP group and Hydrocortisone-treated group. The model of SAP was induced by retrograde injection of 5% sodium taurocholate into the bili-pancreatic duct in SD rats, rats in Hydrocortisone-treated group were given 10 mg/kg Hydrocortisone 10 min after inducing SAP. Rats in the three group were sacrificed at the 3rd,6th and,12th hour after induction of the model.The levels of amylaseTNF-αIL-6TXB_26-Keto-PGF_ 1αTXB_2/6-Keto-PGF_ 1αin plasm, the myeloperoxidase(MPO) in the pancreas and lung, the wet/dry weight ratio of lung were measuredThe histopathological changes of pancreatic and pulmonary tissue were evaluated. Results The levels of amylaseTNF-αIL-6TXB_2TXB_2/6-Keto-PGF_ 1α in plasm, and the MPO in the pancreas and lung significantly increased in SAP group compared with that in the SO group(P0.05). At the 6th12th hour, the wet /dry weight ratio of lung significantly increased in the SAP group compared with that in the SO group(P0.05).Compared with SAP group ,aministration of Hydrocortisone significantly reduced the levels of TNF-αIL-6TXB_2TXB_2/6-Keto-PGF_ 1α in plasm and the MPO in the lung(P0.05), the wet /dry weight ratio of lung also significantly decreased at the 6th12th hour(P0.05), the severity of lung injury was decreased. Conclusion Hydrocortisone may have therapeutic effects on SAP by decreasing the production of TXA_2 TNF-αIL-6.
