Abstract
Objective:To investigate the effect of spironolactone on the expressions of(IL-6and MCP-1)in vascular calcification model of rats.Method:Arterial calcification model was induced by vitamin D3 plus nicotine(VDN)in Sprague-Dawley rats.Calcium contents and alkaline phosphatases(ALP)activity were detected by using calcium assay kit and alkaline phosphatase detection kit respectively.Vascular calcification was determined by Von Kossa staining.IL-6and MCP-1contents in plasma were determined by radioimmunoassay and its receptors immunoactivity were determined by immunohistochemistry.Result:Von Kossa staining showed that mass black granules deposited in aortic wall of the vascular calcified rats induced by VDN.Calcium contents and ALP activity in calcified group were increased significantly than that in control group.The IL-6and MCP-1contents in plasma were increased in calcified group,meanwhile,aortic IL-6and MCP-1receptor levels in calcified group were upregulated significantly compared with that in control group.In addition,spirolactone reduced the degree of vascular calcification,calcium contents,ALP activity,IL-6and MCP-1content.Conclusion:Expressions of IL-6and MCP-1down-regulating by spirolactone is another important mechanism to reduce vascular calcification in rats,maybe.
