Effects of "Spleen-Fortifying, Qi-Regulating and Blood-Activating Compound Recipe"on Plasma Endotoxin Content and TNF-α Protein Expression in Alcoholic Liver Injury Rats

Objective:To investigate the therapeutic effects of Spleen-Fortifying,Qi-Regulating and Blood-Acti-vating Compound Recipe(SQBCR) and its influences on plasma endotoxin content and TNF-α protein expression in alcoholic liver injurg rats.Methods:The model of alcoholic liver injurg was duplicated by feeding Lieber-Decarli liq-uid diet to SD rats.Thirty-two male SD rats were randomly divided into normal control group(n=5),liquid diet con-trol group(n=9),model control group(n=9)and herbs treatmeat group(n=9).Since the 4th week,the rats were admin-istered with the drugs or distilled water respectively,and the specimens were collected at the end of 8th week.The detection indexes included histological changes in liver with HE and oil red O staining method,TG content in liver tissue,liver injury index such as serum ALT and AST,plasma endotoxin content and P-IκB and TNF-α protein ex-pression in liver tissue with Western blot assay.Results:Remarkable fatty degeneration was observed in liver tissues of model rats after feeding of ethanol liquid diet for 8 weeks.Compared with normal control group and liquid diet control group,all the TG content,plasma endotoxin content,serum ALT and AST contents,p-IκB and TNF-α protein expressions in liver tissue increased significantly in the model group.Compared with model group,the fatty degenera-tion and inflammation in liver tissue were alleviated remarkablely and all the other indexes also inproved significantly in SQBCR group.Conclusion:SQBCR has a significant effect against alcoholic liver injurg in rats and the mechanism may relate to its inhibition of plasma endotoxin and protein expressions of p-IκB and TNF-α.
