Abstract

Focused on the issues of explants' effective disinfection schemes in the tissue culture of Aquilaria sinensis,one-year-old seedings of Aquilaria sinensis in the nursery were used as propagation material.Using stem explants, two disinfection experiments with three concentrations of NaClO(5%,10% and 20%) and HgCl2(0.05%,0.1% and 0.2%) were carried out.Based on the result,an orthogonal experiment with L9(34) array of three factors and three levels, namely explants(leaf, terminal bud and stem),NaClO's disinfection time(0,3,5 min) and HgCl2's disinfection time(0,3,5 min),was conducted. The results show the optimal concentration of NaClO and HgCl2 was 10% NaClO and 0.1% HgCl2 respectively.Compared with the explants of leaf and terminal bud, stem explants were much better for disinfection in view of its lower rate of contamination and mortality and relatively higher survive rate. The optimal disinfection schemes was that the stem explants were immersed in alcohol(75%) for 30 seconds, and then were immersed in HgCl2(0.1%) for 5 minutes with the lowest contamination rate(43.33%) and the highest survive rate(31.67%).
