Abstract
Objective To evaluate the suitable method to isolate mesenchymal stem cells from the skin in vitro.Methods Two different methods low serum and serum-free primary culture were adopted to culture mesenchymal stem cells from the skin in vitro.The differences were compared by observing cell morphology under microscope and using flow cytometry techonology.Results Both two methods could culture mature mesenchymal stem cells from skin.But the cultured time,which in low serum group was 8.93 d while in serum-free group was 15.93 d,had statistically significant(P0.001).Compared with serum-free culture method,low serum cells cultured cells with higher positive rate of CD44 and lower negative rate of CD45,which had statistically significant(P0.001).Conclusion Low serum culture is a method to culture skin mesenchymal stem cells,suitable for clinical trial and research.
