Isolation,purification and analysis of polysaccharide of Gynostemma pentaphyllum polysaccharide by IR,GC

Abstract

Objective: To isolate,purify,and identify the structural features of polysaccharide in Gynostemma pentaphyllum(Thunb.) Makino.Methods: The reference to the used of water extraction and alcohol precipitation,DEAE52 cellulose column chromatography and other methods,have refined Gynostemma pentaphyllum polysaccharide(GPM2).Using IR and GC,analysis of polysaccharide structure,monosaccharide composition and molar ratio.Results: The effect of polysaccharide's isolation and purification with DEAE52 cellulose column chromatography is obvious,have refined GPM2.IR showed GPM2 has typical absorption peaks of polysaccharides.GC analysis indicates the monosaccharide composition obtained rhamnose,arabinose,xylose,mannose,glucose,galactose.The molar ratio is 3.23:7.71:1.00:2.29:2.88:14.84,Where the highest concentration is galactose.Conclusion: Analysis the structure and composition of GPM2,has a great direction significance in the function and production of Gynostemma pentaphyllum.
